"To be a scientist requires not only intelligence and curiosity, but passion, patience, creativity, self-sufficiency, and courage. It is not the courage to venture into the unknown...All real scientists exist on the frontier... The best among
them move deep into a wilderness region where they
know almost nothing."
I think that the passage begins with a tone of reverence and admiration for scientists and the contributions they have in society. When he says that scientist "exist on the frontier" and describes how they have to embrace a substantial amount of "confidence" and "intelligence", it almost sounds like he's saying there's a strict guideline to become one.

I think the tone shifts about half way through the article. After he introduces the importance of scientists, he transforms into an aggressive, firm tone. The author becomes very certain in his beliefs. He says, "It is grunt work, tedious work that begins with figuring out what tools one needs and then making them...Experiments do not simply work. Regardless of design and preparation, experiments—especially at the beginning, when one proceeds by intelligent guesswork—rarely yield the 65 results desired." By stating short, solid statements, backed by evidence, the author moves the tone from politeness and respectfulness to solemness.

The author's tone really shapes the feelings that he is trying to evoke throughout the passage. By starting off with a tone of reverence, he can introduce scientists and define their impacts. When shifting the tone in second half, the author transitions to the more solemn tone to create persuasion and a confidence that makes it difficult to disagree.
