How does the secondary metabolite been produced? &nbsp;<br><br>Polyhydroxyalkanoates are a class of natural (not synthetic) polyesters that are derived from bacterial fermentation of sugar or lipids. The production of streptomycin requires continuous fermentation, and the growth of <em>S.griseus</em> requires continuous aeration and agitation. The fermentation process takes 4-10 days. In this phase, the incubation period lasts for 24 hours to 6-7 days. The pH during this phase remains constant between 7.6 and 8. There is little or no production of mycelium.</div><ul><li><strong><em>Third phase</em></strong></li></ul><div>It is the final phase of fermentation, where the concentration of glucose depletes from the medium that results in cessation of streptomycin production, and the bacterial cells begin to lyse. As a result, the medium’s pH increases by the deposition of ammonia after the cell lysis. This acidic water forms through the chemical reaction of surface water (rainwater, snowmelt, pond water) and shallow subsurface water with rocks that contain sulfur-bearing minerals, resulting in sulfuric acid.<br><br>As such, microbes that are able to suvive in acid mine drainage are microbes that uses chemicals as an energy source (chemo-), with its chemical being some type of inorganic chemicals (-litho-) and also uses inorganic carbon, like CO2 (-auto). Hence,&nbsp; microbial community that resides on the acid mine drainage are acidophilic chemolithoautotrophic microbes<br><br>In acid mine drainage :&nbsp;<br><br>-	microbial community: acidophile microbes<br><br>-	nutrient requirement: Sulphur<br><br>-	possible metabolism that are present in an acid mine drainage – chemolithoautotrophy<br><br>-	Sulfate is essentially chemically inert, nonvolatile and not toxic and hence sulfate emissions may be viewed as benign to the environment. However, it is recognized that high sulfate concentrations can negatively influence the natural sulfur cycle.&nbsp;<br><br>-	As a treatment option, Sulphate-Reducing Bacteria (SRB) use sulfate&nbsp; as a terminal electron acceptor while selected organic compounds can donate electrons. This treatment will neutralize the acidity of the drain and also percipitate the heavy metals that present there<br><br>-	Example, the anaerobes of the genus Desulfovibrio are capable of reducing SO to H2S. Hydrogen sulphide is thus promote alkalinity and likely to accumulate in waterlogged soils.&nbsp; Hydrogen sulphide reacts with lead, iron and zinc to form insoluble precipitates. The soil is taken from a mining environment where organic nutrients are extremely low.</div><div><br></div><div>i) Describe and explain the metabolism of this microorganism (8 marks)</div><div><br></div><div>Removal of sulfide minerals is defined as oxidation of sulfide by the microbes. The metabolism of this microorganism is chemolithoautotrophic. This microbe obtains energy from chemical compounds and since the organic nutrients are extremely low, it can derive the energy from the inorganic compound which in this case is sulfide by oxidation of sulfide to sulfate. The oxidation of sulfide can occur in two ways which are aerobically and anaerobically. Aerobically, the oxidation can occur in substrate-level phosphorylation depending on AMP (transfer phosphate group to ADP to produce ATP) or in oxidative phosphorylation (ETC) where the sulfide acts as electron donor and oxygen as electron acceptor to produce ATP. Anaerobically, sulfide will oxidize to sulfate by using nitrate as terminal electron acceptor which then reduces to dinitrogen in ETC with the help of sulfite oxidase enzyme. Since organic nutrients are low, this microbe can only obtain carbon source from inorganic carbon, which is through carbon dioxide fixation (calvin cycle) using the energy source from the oxidation of sulfide. It can also use the energy of the reducing power from reverse electron flow using sulfur as a higher redox potential that can reduce NAD+ to NADH. NADH is then used in the calvin cycle for carbon source. Thus, it is considered as autotroph.&nbsp; Example of bacteria is thiobacillus thiooxidans.&nbsp;<br><br>ii) What is the primary and secondary product of the microorganism? (15 marks)<br><br>Type of location: Sewage sludge<br>Type of microorganisms: Bacteria<br>Species: Clostridium butyricum<br><br>&nbsp; Clostridia are Gram-positive, anaerobic or obligate anaerobic bacteria that are among a metabolically diverse group .Clostridia perform diverse metabolic functions, including the conversion of starch, protein, and purines into organic acids (i.e., acetic, butyric, and caproic acids), alcohols, CO2, and hydrogen. Because of their broad and flexible metabolic capabilities, Clostridium species are included in many microbial co-culture ecosystems, which we will refer to in this review as “Clostridium co-culture systems.”<br>&nbsp; Typically, Clostridium co-culture systems are used to produce biofuels such as hydrogen and CH4, solvents, and organic acids. Because cellulosic materials are commonly found in nature , the specific metabolic capacities of cellulolytic strains and producers in co-culture systems have attracted significant attention and offered many long-term prospects for development. <br>The co-culture of <em>C. cellulovorans</em> and <em>C. acetobutylicum</em> achieved a 2- to 3-fold improvement in hydrogen production in comparison with <em>C. acetobutylicum </em>mono-cultures from lignocellulosic substrates.</div>]]></description>
