-Opened the culture we grew last week.

-Checked for bacterial growth and confirmed that it was H. pylori, not another organism.

-Proceeded with UV induction for the strain, then incubated for one day.

-The next day, centrifuged the sample to obtain the prophage lysate.

-Performed a spot test and waited three days for the results.

2. How is your progress?

   • For the second week, I learnt how to perform UV induction on the H.pylori and it is quite challenging as it needs our hands to be very particular so that we didn't contaminate the strain. Other than that, I also opened the culture that we grew so that we can continue to the next step which is the UV induction. Not just that, I also learnt how to do spot test on the H.pylori to see whether there is zone of inhibition of the growth.

5. ONE good thing from what you did that you can practice now!

   • To be more particular in every thing that we do so that we didn't accidentally cause any contamination especially in this medical field like there is always things that can cause contamination so we as future healthcare workers need to be more particular and careful.

6. Any difficulties/concerns? How did you cope?

• It is not easy to culture H.pylori and there were days when we didn't manage to get any growth and cannot continue to the next step.

• We have to sub back the H.pylori strain and for another 3 days to see whether there is growth or not.

• The only way to cope with that is to be particular in how you culture it so you may not contaminate the agar with other than H.pylori.

• I am getting better at understanding the connection between words and gestures, and I even guessed the correct answer when my teacher asked me about the gesture for certain words

• I need to focus on applying it, not just memorizing and understanding the gestures, because the real purpose of learning a language is to communicate

• I am becoming more curious and excited for the next lesson, and I can’t wait to learn how to apply it and communicate with the deaf community

• I can easily understand the gestures in sign language, which makes it easier for me to learn more and to understand a little when communicating with deaf people, even though I have not fully learned the language.

• My difficulties are still the same, as some gestures look similar to others, which can lead to miscommunication. Therefore, I need to be more careful and precise

• My plan for next week is learning about greetings so it will be more exciting to learn.

2. How is your progress?

During the second week, I learned how to perform UV induction on H. pylori. The process was quite challenging because it required precise handling to avoid contamination. I also opened the culture that we had grown earlier in order to proceed with the UV induction step. In addition, I learned how to conduct a spot test on H. pylori to identify whether a zone of inhibition appeared in the growth.

I feel excited to be gaining deeper knowledge and practical skills in handling H. pylori.

I have learned to be more meticulous in every step, ensuring that I minimize the risk of contamination. This practice is highly relevant to the medical field, where contamination can easily occur, and being particular and careful is essential for safety and accuracy.

Some cultures didn’t grow, so we had to subculture and wait again. I coped by being extra particular during culturing to reduce errors and contamination.

To repeat the procedures I have learned over the past two weeks using different strains in order to collect more data for our research.

11/8 – 2. How is your progress?

- This week I was busier than last week, work comes through my door every minute, I don’t get bored easily as I was really focused on getting my job done before I go back home.

- So far, I really like to do my work even though it is tiring and hard to understand at first.

· Work efficiently and try not to do your work alone and ask your colleagues questions if you do not understand

4. Your feelings and thoughts

· Try to be positive, sometimes I have a lot of free time at my department as they cannot expose the information of company easily to the outsider

· Hopefully as the day goes by, I can learn some of a few things that will help me in the future

5. ONE good thing from what you did that you could practice now!

- Cleaning. I obviously have a really hard time cleaning my room, sometimes it is tidy I don’t have really a lot of time to manage it but from these past weeks every Wednesday I will clean my room as I did at my workplace motivation is not the key, but discipline will always be.

6. Any difficulties/ concerns? How did you cope?

· As far as I am concerned there is none, but I am really annoyed when I need to wake up early in the morning and someone to disturb my workout time. It needs to be fixed. Sleep early. Good food. Less workout time but effective training. Repeat. There it is.

· I try to get involved more in their work and hopefully into their engineering stuff

· Expand my knowledge and get to know job scopes not just at my department but also get to know the other department

This week, I followed the waste disposal process more closely. I got to see how the waste is handled from inside the factory until it reaches the disposal site. I feel like I understand the full process better now.

* Waste from factories is first sorted into different types (e.g. normal, chemical, or dangerous).

* It is stored in special containers to avoid leaks or harm.

Then, trucks come and collect the waste. These trucks use *hydraulic systems** to load and unload the waste.

The waste is then sent to a *treatment or disposal site**, depending on its type.

* Safety and cleanliness are very important in every step.

I felt excited again this week, especially because I could see how everything is connected—from how waste is made, collected, and finally disposed of. It made me realize how important proper waste handling is to protect the environment and health. I also felt proud to be learning something that most people don’t usually see or think about.

I learned the importance of handling waste properly and using protective equipment. I can apply this in hospitals too by being more careful when handling medical waste.

At first, I didn’t really understand the process at the disposal site. There were machines and systems I had never seen before. I asked the workers and they explained it step-by-step. I also took some photos (with permission) and looked up more info later.
