Every year a band of gypsies visits the town, and brings new scientific discoveries. José Arcadio becomes obsessed, and throughout the rest of his life he pursues one theory after another. This also happens to several of his descendants, until the last one unscrambles a prophecy left by a man named Melquíades. It describes each of the events that befall the members of the family. The book ends here, with the town being swept away by "apacalyptic like winds", which simply refer to a hurricane.

The book also follows how Macondo industrializes and grows. Traders set up shops and routes, more and more people pass through, and eventually a banana plantation sets up near the town. This leads to a worker uprising sponsored by one of the great-great grandchildren, and a massacre of the striking workers.
