There then is a conflict between MacBeth and his wife Lady MacBeth who encourages him to take a different course of action that he is hesitant to take.  Once MacBeth stops struggling with his ambition for power the conflict then shifts to the other characters of the story, mainly Banquo and MacDuff, who challenge his authority. 
In this play, MacBeth is both the protangonist and the antagonist. His wife thinks it's a great idea and convinces him to do it. Macbeth decides to go through with it and kills Duncan where he slowly becomes paranoid.
