One aspect of 21st Century learning is being able to collaborate with others. Flexible grouping assists students with developing this skill. If forces them to work in situations that they might not have chosen themselves.

Stations, which are questions or activities taped down on certain desks around the room, are a great way to differentiate. Stations not only allow physical movement within the room (see Rick Wormeli's "Meet Me in the Middle"), but they allow for students to work at their own pace (either individually or within assigned groups). Plan on making more stations than your students can get through within one class period. Some higher-level students will surprise you and complete all stations! Make sure to go over station answers at the end of class. That way, students can reflect and remedy their thought processes. This can serve as an excellent way to review for quizzes and/or tests! I use it in my science classroom, but it is a cross-curricular differentiation strategy.

I like this video's explanation on differentiated instruction and the student's readiness. After following some of my students for a couple years, I'm really noticing that some of them are coming into my classroom with low expectations for themselves and it's been my job to change that; to show them they are capable, and slowly increase the difficulty from there. It's all worth it when I see a student light up when they finally "get it."
It's also been a learning curve for me with students having those gaps as she said. In the past, I would be naive in thinking all my students would have some of the same gaps. Then then along come students who have been stationed in other countries or from home school and have been doing different programs - and they bring their different sets of skills.

Choice Variation is a strategy in which students are practicing the same skills and standards, but submitting the final product in various ways. Choice in the classroom presents students with ownership of their learning, which is crucial for extended learning.
