Bacteria was made from 5 different macromolecules carbohydrates, lipids, proteins, RNA, DNA Avery and his team knew that one of these 5 macromolecules had to be the transforming agent. Avery and his team used enzymes to take away one molecular piece from the bacteria he would then add the rest of the dead S-strain bacteria to a syringe of live R-strain streptococcus he would then inject the bacteria mix into the mouse and wait to see if pneumonia developed. Avery repeated the procedure for the different macromolecules which make bacteria. If transformation still happened he knew he had not taken out the transformation agent. When Avery removed carbohydrates from the dead S-strain and combined it with the dead strain with the live R-strain, injected the mix into the mouse bacterial transformation occurred and the mouse died this told them the transformative agent was still in the mix so Avery repeated this with lipids, proteins and RNA but attained the same result the transformation still happened which leaves one thing DNA, when they removed DNA from the dead S-strain and combined the dead strain with the live R-strain and injected the bacterial mix into the mouse bacterial transformation did not occur and the mouse lived this told them the transformative agent had to be DNA.

They carefully treated this mixture with enzymes that destroyed proteins, lipids, carbohydrates, and some other molecules, including the nucleic acid RNA. Transformations still occurred. Clearly, since those molecules had been destroyed, none of them could have been responsible for transformation. Avery’s team repeated the experiment one more time. This time, they used enzymes that would break down a different nucleic acid—DNA. When they destroyed the DNA in the mixture, transformation did not occur. There was just one possible explanation for these results: DNA was the transforming factor.

Some of these came from the famous “image 51,” a remarkably clear and striking X-ray diffraction image of DNA produced by Franklin and her graduate student. (A modern example of the diffraction pattern produced by DNA is shown above.) To Watson, the X-shaped diffraction pattern of Franklin's image immediately suggested a helical, two-stranded structure for DNA.

Avery and his team put together a mixture of various molecules from the heat-killed bacteria. They carefully treated this mixture with enzymes that destroyed proteins, lipids, carbohydrates, and other molecules including the nucleic acid RNA. Transformation still occurred. Since those molecules were destroyed, none of them could've been responsible for transformation. Avery's team did the experiment again. This times they used enzymes that would break down DNA. After destroying DNA, transformation did not occur. This mean DNA was the transforming factor.
