         <description><![CDATA[<div><strong>I</strong>n this activity, we are extracting total genomic DNA from our collected insects. Total genomic DNA includes DNA, and the symbiotic bacteria if present in the insect.&nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp;<strong>T</strong>he first step is Cell Lysis. We began blotting away the ethanol from our insects. Then, we macerated them with Buffer ATL. This breaks open the cell and cell membranes. the problem with this is that it also exposes DNA to proteins in the insect. The enzyme Proteinase K is added to keep the DNA intact and denature proteins. <strong>T</strong>he second step is the elimination of cellular debris. This is a purification process. We put our tubes in the spin column. This spun or tubes around, and cleansed whatever liquid we had inside. This filters everything other than the DNA. Now that it has been filtered, it was washed by two Buffers: AW1 and AW2. Column washing removes residual protein impurities without affecting the DNA.&nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; <strong>T</strong>he third step is DNA elution. We will complete this step by removing DNA from the filter. This is done by adding the elution buffer AE. Spinning the tube with the DNA in the filter will pull the elution buffer through the matrix, pulling the DNA into the extraction tube. Elution of the bound DNA is typically done with later or a low salt solution like TE Buffer. This is because the removal of salts with the<br>increase in water molecules unmasks charges and rehydrates the DNA as well as the surface of the silica membrane.</div>]]></description>
