3.mengira spesis menggunakan Quadrat 
-jarak 100m diukur dan jarak antara Quadrat 10m. 
-setiap spesis yang terdapat dalam quadrat dikira dan dicatat
4.sampel air di ambil 100ml, kemudian dibahagikan kepada 4 bekas. Bekas pertama untuk mengira oksigen terlarut, bekas kedua untuk amonia, ketiga nitrite dan keempat nitrate menggunakan kaedah hach method.

Method B (Plankton)
1. Bottle is used to collect the water sample from different part.
2. Sample is brought to lab to observe the plankton under microscope.
3. Sample is first observe in petri dish.
4. Water dropper is used to take the sample that shown the plankton from petri dish to add on slide.
5. Observe the sample under the microscope.
