My problem is to come up with a sturdy but light material that will make the backpack durable and easy to handle.

There are three main materials that could possibly fulfill this purpose and solve the purpose. They are Graphene, Self-Healing Plastic, Data-Storing Glass, Permeable Concrete, and Vantablack.

Graphene is probably the most reliable, helpful, durable, and strong material that fits all of the needs of the backpack. It is waterproof, stronger than steel, flexible, durable, versatile, and is two dimensional. This material would help the backpack become completely waterproof.

It could also be used for filtering unneeded microorganisms out of water. The material is also very accesible, comfortable, light, and easily manipulated.There is another kind of graphene that is not waterproof. This feature will filter the water through it but take out all of the dangerous toxins. That is because that type of graphene is completely resistant to any other fluid or particle.
