The affected protein in dogs with curly hair is keratin71. The typical job of these individual keratin proteins is to link together into tough threads that create a web inside cells. The mutation causes the individual keratin proteins to lose the ability to perform their function where no fibers can be formed to support the hair follicles.

A two base substitution in the keratin71 gene changes one amino acid, which alters the shape of the protein. This overall results the misshapen follicle to mold, growing hair into a flat, ribbon-like shape, which makes it curl.

Examples in Other Species

This gene and mutation is present in other species as well. For example, mice with no keratin71 protein have kinked fur, curly whiskers, and they lose patches of fur. Another example is humans. People with that mutation have hair that feels like wool and easily falls out. The lack of keratin proteins causes whatever it builds such as hair, skin, nails, (etc.) weakens it instead.
