She tells Macbeth to act normally and be confident because she fears that people can read his face and see "through" his fear. Macbeth must therefore look like an innocent flower, but be as sinister as a serpent underneath. Lady Macbeth is determined to get the deed done.

Here Lady Macbeth calls upon the "murdering ministers" to assist in the murder of Duncan, just like when she wants to become "unsexed" she wants her inner evil to come through so can become able to kill Ducan. 
The entire quote is very reminiscent of the unaturalness of the witches and just like the witches she wants to become ambigious in her gender. She also calls for darkness in order to cover over her crime because she is perfectly aware that what she is not a noble action. "Nor heaven to peak through the blanket" 
It is interesting that she is not asking the "mudering ministers" for help or for somone else to do it, she is only asking for the personal strength to do what needs to be done.  
This tells us alot about Lady Macbeth. She is uncompromising and seemingly fearless as she gathers her strength. She appears determined to do anything to achieve her goal. 
It also seems clear that she is loyal to Macbeth and is willing to sacrifice her goodness and her gender to achieve this goal, just based on the letter. Lady Macbeth emphasizes the importance of Macbeth’s expression when the king comes to visit. He should look innocent like a flower, but under the surface he should still be sly and devious like a serpent. Under her direction, they will achieve power and mastery of kingship.

These lines show Lady Macbeths ruthless nature and willingness to commit regicide. Furthermore, she wants Macbeth to act like a serpent under the surface and seems sinister in that regard. She seems consumed by the power which it would bring and doesn’t hinder Macbeth from performing an act which they recognize as evil.

Lady Macbeth summons the murderous spirits and calls upon their strengths to murder the king.
She wants them to remove her womanhood and take away tenderness. She wants her blood made thick and to be filled with cruelty. All in all she wants to be cruel and without remorse.
She is delighted over the news of Macbeth's new title. She is sinister because she is planning a regicide.
