Keith Joung, a scientist who studies at Massachusetts General Hospital, says that his lab, as well as other labs, have found off target sites with mutation frequencies ranging from 0.1% to over 60%. He continues saying that low-frequencies could potentially be dangerous if they make the cells grow quicker and lead to cancer. Many other researchers are concerned that changing an entire population could have drastic consequences for the ecosystem too. It could affect predators higher up on the food chain because other pests may emerge. Also researchers are concerned that a guide RNA would be able to mutate over time, so that it targets a different part of the genome.
http://web.b.ebscohost.com/src_ic/pdfviewer/pdfviewer?vid=6&sid=95dcd8e6-00d2-4f13-8abe-52606f2af773%40sessionmgr120&hid=118

How does CRISPR work?

How CRISPR works is almost like going into a book and correcting a mistake, for example if in that novel it said row, row, walk your boat. and you wanted it to say, row, row, row your boat. You would select the word you want to change and correct it so the sentence makes sense. This is exactly how CRISPR work but instead of a book we are talking about DNA. What happens is we are going through DNA and spot a mistake, next we choose to correct it by using CAS9 to first cut the section of DNA and then the section cut is replaced with the correct sequence, correcting the error. (Below is a more detailed guide on how CRISPR works in an article from the Proceeding of the National Academy of Sciences of the United States of America) http://www.jstor.org/stable/23771061?Search=yes&resultItemClick=true&searchText=CRISPR&searchText=testing&searchUri=%2Faction%2FdoBasicSearch%3FQuery%3DCRISPR%2Btesting%2B%26amp%3Bfc%3Doff%26amp%3Bgroup%3Dnone%26amp%3Bacc%3Don%26amp%3Bwc%3Don&seq=2#page_scan_tab_contents

CRISPR is a sensitive topic that is either enhancing normal traits or correcting an abnormality. There is a difference from making a child have green eyes instead of brown and preventing a child from sickle cell anemia. According to an article in Issues in Science and Technology, some people disagree with the use of CRISPR saying that it will eventually change the ways creatures live their lives and their experiences. The think that some life forms will be empowered and others will seem unnatural depending on what procedure took place. Many people think that it will be great for not only humans, but the environment as well, thinking it would improve the world as a whole. http://www.jstor.org/stable/24727002?Search=yes&resultItemClick=true&searchText=dangers&searchText=of&searchText=CRISPR&searchUri=%2Faction%2FdoBasicSearch%3Fwc%3Don%26amp%3Bso%3Drel%26amp%3Bfc%3Doff%26amp%3Bacc%3Don%26amp%3BQuery%3Ddangers%2Bof%2BCRISPR%26amp%3Bhp%3D25%26amp%3Bprq%3Dagainst%2BCRISPR&seq=2#page_scan_tab_contents

The acronym CRISPR stands for Clustered Regularly Interspaced Short Palindromic Repeats. CRISPR is a genetic editing tool that can fix someone's DNA in order to destroy the gene in certain diseases like sickle cell anemia, muscular dystrophy, etc.
http://search.ebscohost.com/login.aspx?direct=true&db=lfh&AN=110929188

Labs have first had to study CRISPR on animals to model human behaviors and success rates. Some of this has been done on lab mice where scientists will inject CRISPR CAS-9 into the embryo of the mouse to fix and alter things, and it has worked! The scientists were able to get different mice and using this knowledge we know that it is possible for human use soon. Now, there has been trials done on a few humans with success and no major side effects. This is a gigantic leap for mankind as a whole.
