- Pharmacological approach in drug target validation involves the use of small molecule inhibitor and monoclonal antibodies to determine whether the cell can survive (diseased cell or normal cell) where monoclonal antibodies bind to unique epitopes but cannot cross cell membranes whereas small molecule inhibitor binds with conserved active site of a target
- Genomics approach in drug target validation uses antisense oligonucleotides, RNA interference (RNAi) and transgenic animals whereby it inhibits or blocks specific gene expression. Antisense oligonucleotides uses RNA complementary to gene of interest that binds to cellular mRNA, preventing translation. RNAi uses cells natural machinery, facilitated by short interfering RNA molecules, to effectively knock down expression of gene of interest. Transgenic animals are bred either with putative gene modified or inactivated which can be used to study drug interactions against potential drug candidates

Pharmacological approach uses small molecule inhibitors which are usually involved in validation of intracellular targets. Monoclonal antibodies are also used in this approach for extracellular targets as the antibodies are unable to enter the cell. Both types of molecules can bind to specific targets and verify their presence and effects in the tested cells.

Genomics approach is utilized through the inhibition of mRNA expression using antisense oligonucleotides. RNA interference can either induce the breakdown of mRNA or block the movement of ribosomes to prohibit translation. Transgenic animals are also used where their genes are modified or inactivated and are then treated with potential drug candidates to study its interactions within the organism.

~Pharmacological approaches are defined as the study of effects of chemical substances known as highly sensitive pharmacological agents that act on living systems of which it includes selectivity testing, pharmacological profiling, testing in animal models of disease and safety pharmacology. These pharmacological agents include small molecules that are not completely specific to target, leading to confirmation bias.

~Genomics approach includes the use of antibodies, ribosomes and siRNA.
Knockout phenotypes are a very useful and powerful approach to decipher gene function and target validation in mammalian physiology.
