In a nut shell, the brain is the center piece of the body that regulates all bodily processes. The three parts of the brain that are affected by drugs are the brain stem, cerebral cortex, and the limbic system. The brain stem controls necessary processes such as breathing, the cerebral cortex controls our senses, and the limbic system regulates the brain's reward system. Drugs slow down all forms of brain activity and when used, it feels "good" therefore creating an addiction in users.
