-This experiment is to test the present of aflatoxin which is toxin produced by a specific mold and glow under black light
-aflatoxin produce by Aspergillus mold.
-this start with aflatoxin test.Then weighing sample,diluting sample and lastly detecting toxin
-in detecting toxin, if the indicator is visible, its means the test is correct.
-the indicator is visible when the middle indicator was dark as bottom
2)BACTERIA SAMPLING.

-In this test, we use 3 different disposable lab equipment
*petrifilm
*disposable pipette
*quick swab
-use two type of milk
i)pasteurized milk
ii)raw milk

-inoculation
>take the first sample using pipette and put on petrifilm
>then incubate at 90F, 32 degree celcius.
>wait for 48 hours(national standard for evaluating milk)
>take out and observed
>results:raw milk contain more bacteria compared to pasteurized milk.

-swabbing
~take from:
   i)tank where milk store before processing
ii)tank where pasteurized milk is store before package

-web swabbing
>tank is dry and need to wet swab method to collect the bacteria

-dry swabbing
, inoculation

-intrepatation
pasteurized tank(13 x 200=260)
milk tank(12 x 20=240)
..this results shown pasteurized tank is contaminated..like pasteurization is not working
-more colonies,more bacteria level

iii) GRAM STAINING
-in this test we use 4 different chemical to stain which are :
>crystal violet~indicate gram positive
>gram iodine~fix the dye to cell wall   
                          of bacteria
                         ~make it permanent
>95%alcohol~wash excess dye 
                       ~leaving only dye fixed
                         bacteria        
    >safranin

-pink radish shown gram negative while purple colour will shown gram positive
-the result fom this experiment is gram positive

ok let continue about the microbiology..from this topic i also learned about the gram stain method....i am waiting for the gram stain lab....i want to  conduct it by myself and observe it under microsope..hahahah.....
gram stain are very important to identify either the microbe are gran (-)ve or gram(+)ve............using microsope is the part that i am waiting for....because it is the symbolic for microbiologist....without this microscope the tiny organusm like virus and bacteria cannot be ssen...this how important the microscope is...

..i want to share about my Steptococcus pyogenes....this bacterium is anaerobic facultative bacterium  and non motile bacterium...it can cause mant type od disease like necrotizing faciitis, scarlet fever and rheumatic fever

before i end my reflective journal for this semester..i want to thank to dr wan and to all my coursemate..all of u are superb and sometime are unpredictable...hahahah
