Gossip: Fleming was not a tidy researcher and usually expected unusual bacterial growths in his culture plates. Fleming had teased Allison of his "excessive tidiness in the laboratory," and Allison rightly attributed such untidiness as the success of Fleming's experiments, and said, "[If] he had been as tidy as he thought I was, he would not have made his two great discoveries."

Discovery of lysozyme: Fleming added nasal mucus to a bacterial contaminated plate and realized that the mucus inhibited the bacterial growth, In the next test, he used bacteria maintained in saline that formed a yellow suspension. Within two minutes of adding fresh mucus, the yellow saline turned completely clear. He extended his tests using tears, sputum, cartilage, blood, semen, ovarian cyst fluid, pus, and egg white showed that the bactericidal agent was present in all of these, except on the tears treatment. Fleming wrote: "I wish to draw attention to a substance present in the tissues and secretions of the body, which is capable of rapidly dissolving certain bacteria. As this substance has properties akin to those of ferments I have called it a "Lysozyme," and shall refer to it by this name throughout the communication.

In 1863 some industries producing alcoholic beverages in France were having problems with contamination during the fermentation of wine, which is why Napoleon III decided to bring the problem to Pasteur. Pasteur realised that the contamination problem was due to the presence of certain harmful microbes, so he came up with the idea of heating the wine to a temperature of around 60°C in order to eradicate these pathogens. Years later, this process would be called pasteurisation in honour of Louis Pasteur, its discoverer.

Interesting fact: At the age of 30, he was decorated with France's highest award: the Legion of Honour, an order created by Napoleon Bonaparte in 1804.
Fun fact: During his childhood he wanted to be an art teacher and during his university studies, his grades in chemistry were "mediocre".

Bibliography
Latour, B., 1993.

Lynn Margulis is known due to her discovery about the origin of the eukaryotic cells. In her article "On the origin of the mitosing cells" Margulis proposed that a archaea engulfed an alphaproteobacteria giving origin to the mitochondrion; next, this proteocell engulfed a cyanobacteria giving origin to the chloroplasts. The first step of this theory came up by the thought of that those organelles had their own DNA. In adittion, this thoery is very important because it explains the origin of aerobian life in the earth; also, it explains the origin of flagelum. It was a very hard process because her theory was rejected by the neodarwinism due to the idea that evolution only happens by mutations.

Fun fact: Her paper ("On the origin of the mitosing cells") was rejected 15 times by many journals!

Referencias:
[1]J.

Born on December 27, 1822 in France, Louis Pasteur was one of the most important founders os medical microbiology, he was a French chemist and microbiologist. He pioneered the study of molecular asymmetry, discovered that microorganisims cause fermentation and disease. Was the first to originated the process of pausteurization. In 1843 Pasteur was admitted to the École Normale Supérieure. In 1854 Pasteur was profesor of chemistry and dean of the science faculty at the University of Lille, while he was working there he helped solve problems related to alcohol production at a local distillery and this began the studies on alcoholic fermentation. Also, Pasteur investigated a range of aspects in fermentation including lactic acid that is responsable for souring mil and studied butyric acid fermentation. This helped rise the germ theory of fermentation.

Reference:

Ullmann, A. (2022, May 2). Louis Pasteur.

Born in 1853 in Copenhagen, Denmark, he studied botany at the University of Copenhagen and his study introduced him to the basics of pharmacology and the use of the microscope. After graduating from medical school in 1883, he settled in Berlin.

In 1884, while working with infected lung tissue from cadavers the staining technique was developed in which it was possible to observe bacteria in lung tissues [1]. This procedure consisted of drying a liquid smear on a glass slide over the flame of a burner and adding gentian violet. After rinsing the slide with water, Lugol's solution (potassium iodide) was added, which was assigned as a fixative for the dye and, finally, it was washed with ethanol.

It emerged that after washing with the solvent that some bacteria retained the color (grampositive) while others discolored after this step (gramnegative)[2]. These colorations are due to the cell wall, for the gram positives that will remain purple they have a single thick cell wall and the gram negative bacteria are discolored because they have cell walls with much thinner layers that allow the solvent to remove the dye.

Finally, a final step was added by the German pathologist Carl Weigert (1845-1904), which consists of performing a safranin counterstain to identify gram negative bacteria [3].

Bibliography
[1] P. (Belles X. 2008) (EcuRed, 2015)

Lazzaro Spallanzani: Characterized by his contributions to experimentally refuting the spontaneous generation; defended by contemporaries such as Turbeville Needham. (Belles X. 2008)

• Interesting facts and gossip

Turbeville Needham: Once Lazzaro Spallanzani 1769 succeeded in refuting his idea of spontaneous generation, Needham continued to argue that the presence of air was necessary for the spontaneous generation of microorganisms, but Spallanzani's experiments excluded this factor. He mentioned that some infusions of microorganisms resulted from certain kinds of ´seeds´ that produced some organisms like themselves, which were present in the air. (Doetsch R N., 1976).

Lazzaro Spallanzani: In addition, Lazzaro Spallanzani brought considerable contributions to the regenerative capacity of specific organisms, blood circulation and characterized gastric juices, finding that they were acidic pH and had digestive functions, assessed the role of sperm in reproduction, echolocation in bats, resuscitation of resistance forms of rotifers, nematodes and tardigrades in sand or molluscs, contributions to human respiration with the ability to convert oxygen to carbon dioxide, and experiments with insects and larvae. (Belles X. 2008)

• How he made his discovery

John Turbeville Needham did this by heating containers with beef and vegetable stock, covering them with cork stoppers – which were not covered up properly. Needham would assume that, by boiling the broth, it would succeed in eliminating all the types of microorganisms present there. However, after a few days, it showed the presence of microorganisms in the broths, concluding that the organisms had developed in the broth. Likewise, in 1745, he put to boil some pieces of meat, again with the idea that this would kill all the microorganisms present in that food and proceeded to put it in a container that was not adequately sealed. He did this because he considered that he required the air for the experiment to occur correctly. After a while, he evidenced the formation of microbial colonies on the surface of the food and concluded that these were spontaneously caused by non-living matter (Belles X. 2008) (Ecured, 2015).

Lazzaro Spallanzani: One of the most recognized experiments was refuting spontaneous generation theory experimentally. Spallanzani repeated the procedure used by Needham; however, he made a slight modification to one of the procedures, which would consist of sealing the vessels with more outstanding care; this was how he demonstrated that the broths did not generate microorganisms spontaneously.

Antoine Van Leeuwenhoek was the biologist who first discover microscopic life. He was a trader who used to put the fabric that he traded under the lens of the microscopes. His curiosity about this element lead him to his real passion: the fabrication of magnifying glasses and what he could find looking through it. In consequence, he created a microscope with a 300x lens in which he could look to microscopic objects. At this moment, Van Leeuwenhoek life was focused on looking different objects through the lens. For example, he analyzed moldy bread, the tartar of his theeth, his blood and semen discovering fungus, bacteria, red blood cells and sperm in motion respectively. Therefore, he discovered how protozoa look like by experimenting with pepper and rain water rested for days because of his curiosity about the itchiness caused by pepper. For all his investigations in the microscope Van Leeuwenhoek owns the name of 'the father of microbiology'.

Interesting Fact: Van Leeuwenhoek’s investigations were an important part of the end of beliefs as that semen contained mini-babies and that fleas came from sand. But it took almost a century for someone to continue his investigations.

References:
Doménech, Francisco (2018, August 24). El comerciante que descubrió la vida microscópica. He was born in 1928 (New York), his academic career started with a physics degree in 1950 which led him to pursue a biophysics doctorate that he finished in 1953.
He is known for proposing the phylogenetic structure for life in three domains: Bacteria, Archaea and Eukarya. In 1977 he had sequenced 16s rRNA from multiple species of bacteria and 18S rRNA from eukaryotes. When the sequences were compared between each other, the results indicated that the organisms could be divided in three groups. There was when he realized that the prokaryotes were not just one group but two: Bacteria and Archaea. The scientific community did not like his theory, and it was not until 1987 that he proposed the first phylogenetic tree dividing the three domains.
Because of his discovery, he won a Crafoord Prize (he did not win a nobel because Alfred Nobel hated biologists or so).

Discovery: first vaccine

Edward Jenner was an English scientist and surgeon born on May 17, 1749, in the United Kingdom and died on January 26, 1823, in his native country. In 1775, Jenner made a detailed study of the relationship between cowpox and human smallpox[1]. After testing on animals, he discovered that if the extract was taken from a smallpox wound and injected into a person, that person was protected against smallpox. In 1796, he transplants his first patient, James Phipps, an 8-year-old boy, who took the substance from the hands of a milkmaid who had been infected, it is discovered that James has cowpox. A few days later, the boy was vaccinated against smallpox. As expected, the boy did not contract the human version of smallpox. Smallpox virus cultivation has provided some degree of protection against smallpox[1].
He presented his research by describing his experiences, but Jenner couldn't explain why this method worked, because the virus couldn't be seen under a microscope. The scientists reacted by saying that his idea was too revolutionary and that they needed more proof. Jenner had witnessed many other children, including his 11-month-old son. In 1798, his research was published in 1798, when the term "vaccine" was invented from the Latin "cow". His critics, especially the clergy, considered it repugnant and disrespectful to plant a person in material from a diseased animal. However, the benefits of vaccination and the protection it provides have been demonstrated, and the practice has become widespread[1].

References:
[1]BBC News Mundo.

REFERENCIAS
[1] J.

Cohn is considered the father of bacteriology; one of his major contributions was the discovery of the formation and germination of resistance spores or endospores in bacteria, particularly in Bacillus subtilis [1]. In addition, he managed to note their resistance to high temperatures; from this, he refutes the theory of spontaneous generation [1]. He did this by means of "completely boiled flasks of infusions of hay and Navo cheese" [1]; from this experimentation he speculated that the rapid reappearance of bacteria was due to the formation of heat-resistant spores, and thus proved the fallacy that all bacteria died in infusions [1].
A curious fact about Cohn is that he obtained his Ph.D. at the University of Berlin when he was only 19 years old [2].

REFERENCIAS
Otero, P.

Anton van Leeuwenhoek was born in Delf-Holland in 1632, he had a hobby of carving his own lenses from ordinary glass. Anton participated in the creation of the first scientific society The Royal Society of London, inspired by the discoveries of Hooke, he designed a very small lens of 3-4mm in diameter that could magnify 10 times the original image which would be considered in 1675 as "the first simple microscope and by this he observed the first mobile living beings that were not seen with the naked eye" (Restrepo, 2012). He discovered protozoa in samples of stagnant water and in 1677 recorded the discovery of spermatozoa. In general, it can be said that Anton van Leeuwenhoek "opened for science a door to the microscopic world" (Restrepo, 2012).

REFERENCIAS
Restrepo, M. (2012).

This was done by the Reed team, led by the American physician Walter Reed in Cuba (Crosby, 2007).

Interesting facts:

- One of the Reed team members were possibly self-inoculated or accidentally bitten by the mosquito being studied and died as a result (Suárez & Ordoñez, 2010).

- They worked with volunteers and were paid in U.S. gold (Suárez & Ordoñez, 2010).

-The Reed Commission experiments were considered by Henry R. This team began its work, evaluating Sanarelli's position, by performing an autopsy on 11 patients who died of this disease, without finding evidence of B. icteroides in blood, liver, spleen, kidney, gallbladder or small intestine cultures. When this method failed, they used mosquitoes and injections with blood from patients with active disease. Thus, by the end of 1900 the team had demonstrated unequivocally that the Aedes mosquito was the vector of yellow fever and, in addition, had shown that the disease was transmitted by blood transfusions (Suárez & Ordoñez, 2010).

References

Crosby, M.

1. Antoni van Leeuwenhoek, el primer microbiólogo. The most interesting thing about all his story is the way he decided to tell the word about his discoveries, he sends a letter to the Royal society of London, in which he apologized in advance if he could scandalize society for his findings, especially for his discovery with sperm, allowing the society to decide whether to publish their discoveries or destroy them if they thought it was appropriate. Among his most important discoveries are, demonstrating how red blood cells circulated through certain parts of animals. He discovered the animalcules as he called the protozoa and bacteria in rain and human saliva. The discovery of spermatozoa and humans and animals. He categorized bacteria into three types, bacilli, cocci, and spirilla, and finally opposed the theory of spontaneous generation, stating that weevils hatched from eggs.

Bibliographical references:

1. Leeuwenhoek and Microscopic Life.

Robert Koch contributed significantly to microbiology (specifically bacteriology) in the time when the most important finds where in the area of bacterial cultures. His most important invention was the introduccion to the solid medium and procedure for the obtention of cultures of pure species through the use of grooves in solid medium. This technique was crucial in the big evolution of the microbiology during the last decades of the XIX century. This method constist in the use of a solid medium (usually agar) for the growth of bacteria inoculating this with bacteria achieving pure colonies from the paternal cell. A fun fact is that the first solid bacteria culture medium that he used was a potato slice that he sterilized with a hot knife and protected with a glass cap from air contamination.

References
- Collard, Patrick. He was sent to Amsterdam to become an apprentice to a linen draper. He returned to Delft when he was 20 and established as a draper and haberdasher. Although Leeuwenhoek’s studies lacked the organization of formal scientific research, his powers of careful observation enabled him to make discoveries of fundamental importance.

Discovery: in 1674 he likely observed protozoa for the first time and several years later bacteria, and calculated their sizes. In 1677 he described for the first time the spermatozoa from insects, dogs, and humans. In 1680 he noticed that yeasts consist of minute globular particles. He also gave the first accurate description of red blood cells.

Disturbing fact: in his first marriage, before his wife died in 1666, the couple had five children, only one of whom survived childhood.

Sources:
1) Britannica, The Editors of Encyclopaedia.

María Fernanda Herreño Barbosa - 201912864
Elizabeth Bugie Gregory was an American biochemist (Macho, M. 2019) and microbiologist (Eveleigh, D. Et al. 2018) who was born on October 5, 1920 and died on April 10, 2001 (Angelova, L. 2018). She participated in the discovery of streptomycin, an antibiotic that acts against Mycobacterium tuberculosis (Macho, M. 2019), a bacterium that causes tuberculosis, a disease that attacks the lungs and other organs (CDC. 2016).

This scientist made this important discovery when, after graduating, she began to study how different agents act against Mycobacterium tuberculosis (Eveleigh, D. Et al. 2018) in such a way that, during her master's degree, in company with Albert Schatz served as the "central author" or second author (Angelova, L. 2018) of the article that "announced the discovery of streptomycin" (Eveleigh, D. Et al. 2018). However, and despite all of the above, sometime later, Bugie's colleagues patented her discovery, excluding her from it and removing the credits.
Important data:
- According to statements made by the daughters of this scientist, Burgie said that at some point her colleagues approached her to tell her that "one day she would get married and have a family, so it was not important that her name be found in The patent."
- Bugie's colleagues then started a lawsuit regarding who was truly responsible for the discovery.
- Waksman received a Nobel Prize in medicine in 1952 for the discovery of streptomycin. (Oñate, C. 2020)
- According to Wasksman and Schatz, Bugie only played the role of an assistant researcher, who was in charge of corroborating the results (Angelova, L. 2018)
- Bugie received a small percentage of the monetary profits from the streptomycin patent, which, it should be noted, was much less than what Wasksman and Schatz received. (Angelova, L. 2018)
- Bugie also carried out many more research works that have greatly helped the development of microbiology.

REFERENCE:
- Macho, M. He made this invention when he was consulted because the wine bottles were being damaged in transport. Pasteur realized that this was due to contamination, but it couldn't be boiled because it would damage the taste. Later, Pasteur discovered that simply heating the wine to 55º C killed the bacteria without ruining the taste. A curious fact it that when I was 45, Pasteur suffered a stroke that partially paralyzed his left side, then his partners set up a mobile laboratory so he could still working on his sickbed.

References
(2015). This is a very important technique that “amplify, or copy, small segments of genetic material” (NIH. 2022), in specific, it “amplifies specific fragment of DNA molecules” (Chen, S. El al. 2022) and it is applied in the medicine, genetics (Chen, S. After that, in 1985 he explained the PCR principles in a publication, and later in 1987 the PCR was patented. (Shampo, M.

Jacques Monod devoted himself to his research at the Pasteur Institute, where he established a fruitful collaboration with another war hero, Francois Jacob. In 1959, with the help of Arthur Pardee, they conducted one of the most famous experiments in the history of modern science: the “PaJaMo”, which allowed them to discover the existence of genes regulating the expression of proteins, using a control mechanism (the “Operon Model”). The key to this model was a repressor molecule that can bind to DNA, and whose presence or absence —determined by a regulatory gene— blocks or allows the production of a protein. It is one of the basic mechanisms for life, and in fact this functioning is common to the cells of all living beings (Barral, 2020).

Curious fact:

In 1936, Jacques Monod was about to embark on a scientific expedition to Greenland when one of his colleagues from the faculty of natural sciences at the Sorbonne, Boris Ephrussi, convinced him to accompany him to the California Institute of Technology (Caltech) in the USA to study genetics on a Rockefeller scholarship. That decision saved his life, as the ship on which he was to travel with the expedition sank and there were no survivors (Barral, 2020).

References:
Barral, M.
