As more sectors of your drive are filled up, there is less storage for newer applications and processes to use. This can also be especially apparent if you're running low on memory, where parts of your storage will also be used as swap.

So, how does Disk Cleanup help?
Disk Clean-up helps by searching your drive files that are either no longer in use, temporary or generally just unnecessary. However, if you notice that your system is starting to feel a little slow and can see that the storage has filled up considerably, it'd always be a good place to start to run disk clean-up so that you can clear up any un-used space in your drive.

Therefore if this is the case, clearing un-necessary files can definitely improve your runtime performance by either ensuring that there is enough temporary storage for your programs to use which isn't stored in the RAM, or it will ensure that if your RAM does happen to fill up, there is enough swap memory available to prevent your system from crashing, which is always a good thing.

Another effect is that you may have noticably more personal storage available. If your drive was filling up, you may not have enough room in the future to save pictures, music or any other personal documents, which you will now be able to avoid by having that additional storage space.
