"O this woodcock, what an ass it is!" (1.2.161) Grumio says this after Gremio's claim about "learning." This quote is funny because Grumio is indirectly making fun of Gremio pretending to be scholarly, and is followed by Petruchio and Hortensio essentially telling him to shut up.

-This is when Katherine and Hortensio are fighting, and Hortensio makes fun of Katherine for having no friends. It is funny because she says her mates and he corrects her that she has none.

Baptista: " O, O, Pertuchio! Tranio hits you now." (5.2.57-59)

This is funny because Baptista is basically going "Ohhhh burn!" but in Elizabethan form in response to Tranio's remark.
