Merchants and empires across Asia, Africa, and Europe benefited from this trade route. The Roman and Ottoman empires, the Abbasid Caliphate, and the Venetian Republic significantly profited from the Spice Routes, because they controlled the trade, acted as middlemen, established monopiles (control of a supply or trade in a commodity), and set rules for vital sea routes. This led to increased wealth and power. In addition to amassing imported goods, diplomatic relationships were also formed between the East and West.

In spice-producing locations such as the Banda Islands, Indigenous populations suffered from forced displacement, enslavement, and violence from European colonial powers that were seeking to control the trade routes. Middle Eastern and Ottoman merchants were also impacted as Portugal and other European nations bypassed established routes and Muslim-controlled territories, causing economic disruption and loss of monopoly. The spice trade contributed to a rivalry between European powers causing violent conflicts, corruption, and debt.

The Portuguese Empire fractured the Arab and old Venetian control over the spice trade by pioneering a direct maritime route to Asia. Thanks to Portuguese explorer Vasco Da Gama, in 1498, a new ocean route was devised, and naval power was used to seize control of key trading ports in the Indian Ocean. This established a monopoly by the Portuguese in the 16th century.
