Cannabis use is associated with neuroanatomic alterations in brain regions rich in cannabinoid receptors, such as the hippocampus, prefrontal cortex, amygdala, and cerebellum. The same review found that greater dose of marijuana and earlier age at onset of use were also associated with such alterations.

Cardiovascular
There is serious suspicion among cardiologists, spurring research but falling short of definitive proof, that cannabis use has the potential to contribute to cardiovascular disease.[83] Cannabis is believed to be an aggravating factor in rare cases of arteritis, a serious condition that in some cases leads to amputation.

Cancer

As of 2015, there is no consensus regarding whether cannabis smoking is associated with an increased risk of cancer

Toxicity
THC, the principal psychoactive constituent of the cannabis plant, has low toxicity. The dose of THC needed to kill 50% of tested rodents is extremely high.
