  • Got a leaf from top and bottom of a plant.
  • Traced leaves on graph paper.
  • Found surface area of each leaf by counting the number of boxes covered on graph paper.
  • Put a thin strip of nail polish on the top of both the leaves.
  • Once the nail polish was dried, place a piece of tape on the nail polish.
  • Put the tape sticky side down on the slides.
  • Put bottom leaf slide in microscope on low power and medium power.
  • Observed the slide with just my eye then with the microscope.
  • Estimate how many open and closed stomata were on the slide.
  • Calculate stomatal density: Divide the number of stomata by the surface area of the leaf.
  • Repeat 7 through 10 for top leaf.
