Catalase is an enzyme used by the liver to decompose the poisonous hydrogen peroxide (H2O2) into oxygen (O2) and water (H2O). This experiment seeks to prove that catalase is the cause of this reaction and that oxygen and water are the byproducts, and also to explore the effects of increased temperature on catalase activity, and enzyme activity in general.

Materials

- 5 Test Tubes
- (at least) 5 Liver Pieces
- Hydrogen peroxide (H2O2)
- Water (H2O)
- Wooden Splints (to light a fire)
- A Burner

Procedure

1. Label the test tubes with numbers from 1 to 5.
2. Fill test tube 1 to the 2cm mark with water.
3. Drop a liver piece into test tube 1. Use a splint to push it down if necessary.
4. Record your observations.
5. Fill test tube 2 to the 2cm mark with hydrogen peroxide.
6. Drop a liver piece into test tube 2. Use a splint to push it down if necessary.
7. Record your observations.
8. Fill test tube 3 to the 2cm mark with hydrogen peroxide.
9. Light a wooden splint on fire and wait until the splint cools to a red hot glow.
10. Place the splint in the peroxide.
11. Record your observations.
12. Fill test tube 4 to the 2cm mark with hydrogen peroxide.
13. Drop a liver piece into test tube 4. Use a splint to push it down if necessary.
14. Light a wooden splint on fire and wait until the splint cools to a red hot glow.
15. Touch the splint to the substance produced by the liver-peroxide reaction.
16. Record your observations.
17. Drop a liver piece into test tube 5. Use a splint to push it down if necessary.

18. Heat test tube 5 with the burner.
19. Fill test tube 5 to the 2cm mark with hydrogen peroxide.
20. Record your observations.

1. Enzymes are proteins that act as catalysts for organic reactions. Enzymes lower the energy required to trigger a chemical reaction, resulting in biological reactions and processes occurring more quickly, and allowing for complex life forms to sustain themselves. Enzymes contain a cavity called the active site, to which the substrate, the substance whose reaction is catalyzed by the enzyme, bonds, forming the enzyme-substrate complex. Only one type of substrate can bond with the enzyme, in a lock-and-key manner. The enzyme then breaks the substrate down to form the products of the organic reaction. The enzyme remains unchanged throughout the reaction, and can thus be reused.

2. When hydrogen peroxide (H2O2) was added to the liver in test tube 2, the catalase in the river bonded with the peroxide (the substrate), forming the catalase-peroxide complex. The catalase then broke the peroxide down into water (H2O), and oxygen (O2). The water was the liquid that remained in the test tube, while the oxygen was the gas that escaped through the bubbles.

3. The burning wood splint used in test tube 4 is used to prove that oxygen is being produced in the catalase-peroxide reaction. Since peroxide by itself is not flammable, as was proven by test tube 3, the flame produced by the wood splint touching the bubbles must be produced by a flammable byproduct of the reaction. The only flammable byproduct the reaction could produce is oxygen (O2), which means that oxygen is the cause of the flame and the byproduct causing the bubbling.

4. An increase in temperature denatures the enzyme, as is seen with the catalase in test tube 5. After being heated, the active site of the catalase has changed shape, and the peroxide can no longer react with it. This results in very few bubbles in test tube 5.

In this experiment, it was identified that catalase, an enzyme in the liver, breaks hydrogen peroxide down into oxygen and water. Catalase was identified to be the cause of the reaction, because, without the liver, no reaction occurred. Oxygen and water were also identified as the byproducts, by showing that the inflammable hydrogen peroxide became flammable oxygen gas, which re-lit the wooden splint, and water, which put the splint out. This reaction was also identified to be specific to peroxide since this reaction did not occur when the liver was placed in water. Lastly, it was shown that significantly increasing the temperature of catalase results in little to no reaction, or enzyme activity. It was concluded that this can be generalized to all enzymes.
