Il est le fondateur de la Sorbonne, établissement créé pour permettre à des étudiants séculiers en théologie d'étudier sans être en prise à des difficultés matérielles. « Vivre en bonne société, collégialement, moralement et studieusement ».

Elle devint la Sorbonne lorsque Robert de Sorbon, le confesseur du roi, lui donna son nom. Après 1885, la Sorbonne devint la plus importante université de France ; et, aujourd'hui, elle est toujours l'une des principales universités de Paris. L'église de la Sorbonne fut construite de 1635 à 1642 par Jacques Lemercier, suivant la volonté de Richelieu. Le tombeau de celui-ci se trouve par ailleurs dans l'église, sculpté par François Girardon en 1694. La coupole est l’œuvre de Philippe de Champaigne.
