There are 3 main steps to PCR

1. Denaturation: In this step the two strands of DNA are heated up causing the strands to separate.

2:Annealing: Primer (which is a short sequence of DNA) binds to the DNA strand and causes Polymerisation, in this process free nucleotide form to make a strand of DNA.

3:Extension: The original strands are used as templates for new strands. Firstly DNA polymease joins the free DNA nucleotide together. Order is determined by the original strands sequence of nucleotide. The result of PCR are two double stranded sequences of the DNA, both containing 1 strand of the original strand and 1 new strand.

This process is then repeated many times and in 2-3 hours a billion copies can be made.

Gel Electrophoresis begins with loading the gel. The gel is made out of a polysaccharide called agarose. The gel allows the DNA molecules to travel within in. At one end of the electrophoresis tank there are well, where the DNA will be placed.
Once the gel has been loaded into the gel box, the DNA samples are loaded into the wells. Next the power is turned on and the fragments travel through the gel.
By the end, the fragments should now be separated by size. The fragments that traveled the farthest are the small fragments and the large fragments will remain close to the wells.
Once the fragments have been separated, the gel can be examined and compare the size of the bands. Bands is a defined line of DNA on gel. Bands contain several DNA fragments. The gel can be stained with dye and put under a UV light, indicating the DNA present along the gel. The single fragment DNA will not appear on the gel.
By utilizing a DNA ladder, we can compare the bands to determine its approximate size.

DNA is retrieved from the nucleus and is known as Genomic DNA.
