One of the theories with enough evidence to back it up is that a large meteor hit Earth. While the impact probably did a lot of damage to the ecosystem, the impact alone wouldn't cause mass extinction. That is where the second part of the theory comes into play. The idea that leads to mass extinction is that the meteor created so much gas (from impact and fire due to speed of meteor) was emitted that it blocked out the sun, killing most plants. Without plants, the ecosystem was wiped out.

He said that continents moved across the Earth over time, which explains why continents have similar shapes with each other and why the same species of fossils were found in different continents that are whole oceans apart. This was later disproved, but helped create the tectonic plate theory after studying the magnetic fields from the North Pole shifting (which either meant the North Pole - something in a fixed point - was moving, or tectonic plates were).
