On the basis of the literature review, we used EU(III) chelate 120 times more compared to antibody concentration. To calculate the correct concentration of the chelate we measudred the fluorocence of the chelate-antibody mixture with Arcus 1230 fluorometer (Wallac). After the sample preparation we balanced the FPLC-column overnight.

In order to learn to program Arduino Uno, we completed some practice exercises. To determine if the device and the components were working, we created a code that caused the LED F L I C K E R at one-second interval. To further test our Arduino, we made a second test where we measured the temperature of our hands. After the tests were done, in light of these expected findings, we determined that the equipment we used was working.

in order to be introduced to Arduino we made exercises. We studied electric circuits and biochemistry of the glucose test in order to understand the test and it’s different components. After we understood how the test works, we began practicing programming of our device by tinkering.

After the activation reaction, the extra activation reagents of the sample were washed and the solution was centrifuged (10 min + 10 min, 20238 g). After centrifugation the supernatant was removed and the pellet was dissolved into the conjugation buffer again. The sample was then sonicated for 3 minutes and vortexed between the sonications.

In order to understand the PCR work better we made primers with computer programs. To determine the best primers we solved their melting temperatures an GC concentration with oligo analyzer. The primers were meant to be specific to MecA gene and not bind to other genes. We used Eu-Gene program and clustalW algorithm to design the primers. We calculated the volumes of compounds in our reaction mix and proceeded to design the PCR program.

In an effort to evaluate the presence of mecC and mecA in each sample we designed primers for them both. In order to build the optimal 3´- and 5´-primers for mecC and mecA, we used NCBI gene data bank to find the sequences of both genes and to rule out mismatches, which was DNA from human and E. coli. Ugene was used to determine the melting temperature of the primers and the suitability for both genes. The designing was done for the sake of practice, and instead of these primers, we used pre ordered primers in this study.
