Everyone just discovered that Duncan has been killed and Macbeth killed Duncan's guards, this is when Duncan's sons also run away. These exact lines reveal that the sons left because they felt threatened due to their father's sudden death. But since this was an aside, the other characters do not know this information, making the two sons look suspicious and guilty to their peers.

It's a example of dramatic irony since we the readers/listeners know that the King's sons fled Scotland out of fear of who did murder their father and for their safety, but Macduff and Ross don't know that.
