The position, tone and intensity of the light are really important factors to keep in mind when trying tell a story successfully. 

Three point lighting is a technique used to mimic natural lighting (sunlight) which is what our eyes are most used to seeing. This does not mean that we can't use this technique to create diferente types of lighting that aren't natural.
First of all we set up the Key light, this is usually placed to one side of the subject at an angle and is usually the brightest light . This will illuminate one side of the subjects face and will create harsh shadows on the other side.
Then we have the fill light. This will be placed on the opposite side of the key light and at a lower intensity to soften out the shadows from the fill.

When chosing colours you must always consider the hue, saturation and value of the colour

Colour can be used in many ways, one of the is to show character development, like in the film "The Lorax" it is very colourful and bright, the main character the "Once-ler" appears in very dull grey colours, but as he develops throughout the film and becomes more greedy his clothes become green. 

Colour can also be used to set the mood of scene or an emotion of a character for example in the film "Alice in Wonderland" the colour blue has been used to set the scene and make it look like it was shot at night. Whereas in the Harry Potter the blue has been used to represent Snape's sadness for Lily's death.
