The meaning of this quote is that God's judgment can be terrifying and if you are condemned to hell you would live your life in suffering. Edward views it as stupidity if you dare go against God's word. It relates to the theme because he speaks about God, the judge of the afterlife, when being judges you simply be "born again" in a way, or you could be sent to hell to repent for all the sins that you've committed.

The meaning behind this quote is that even if you were to die, the suffering wouldn't end, and it would follow you to the afterlife. Edward uses a bitter tone when talking about this to demonstrate his beliefs. This quote relates to the being "born again" theme because Edward says, "no end to this exquisite horrible misery." which seems like he is indirectly speaking of the after life; being born again.
